Methotrexate Toxicity: Molecular Mechanisms and Management

نویسندگان

چکیده

Methotrexate (MTX) is the most widely used drug in cancer chemotherapy and considered to be first-line for treatment of a number rheumatic non-rheumatic disorders. The pulmonary toxicity, hepatotoxicity MTX are two its major side effects. Other toxicities such as endocrinological GI cutaneous hematological fatal malfunction or loss, malignancy can also occur, but at significantly lower rate prevalence. This review aims provide comprehensive understanding molecular mechanisms methotrexate toxic effects Lastly, we discussed management this toxicity.

Molecular Mechanisms of Pesticide Toxicity

The environment represents a key contributor to human health and disease. Exposure to many environmental stressors such as pesticides have detrimental effects on health and are considered to contribute substantially to most diseases of major public health significance. Pesticide toxicity has been clearly demonstrated to alter a variety of physiological functions. In addition, evidence suggests ...

Molecular mechanisms of fluoride toxicity.

Halfway through the twentieth century, fluoride piqued the interest of toxicologists due to its deleterious effects at high concentrations in human populations suffering from fluorosis and in in vivo experimental models. Until the 1990s, the toxicity of fluoride was largely ignored due to its "good reputation" for preventing caries via topical application and in dental toothpastes. However, in ...

Evaluation, Identification, and Management of Acute Methotrexate Toxicity in High-dose Methotrexate Administration in Hematologic Malignancies

The pharmacological and medical management of complex chemotherapy regimens are vast and complex, requiring an intimate understanding of physiology, particularly when novel biologic agents are utilized with commonly used regimens. The molecular classification in patients with diffuse large B-cell lymphoma (DLBCL) is multifaceted, particularly with the expansion of novel molecular targets. The p...

Mechanisms, manifestations, and management of digoxin toxicity

Well known functional structures are implicated in the development of digoxin toxicity in the heart: (Naþ/Kþ-) Mg2þ-ATPase, the Naþ–Ca2þ exchanger, and sarcoplasmic reticulum; there is both direct and indirect involvement of sodium, potassium, and calcium ions. The therapeutic effect of digoxin in doses between 1 and 2 ng/ml involves the alpha 2 isoform of Naþ/Kþ-ATPase. Toxic effects occur at ...

Molecular mechanisms of amyloid oligomers toxicity.

Amyloid oligomers have emerged as the most toxic species of amyloid-β (Aβ). This hypothesis might explain the lack of correlation between amyloid plaques and memory impairment or cellular dysfunction. However, despite the numerous published research articles supporting the critical role Aβ oligomers in synaptic dysfunction and cell death, the exact definition and mechanism of amyloid oligomers ...
